Primary neuronal cultures stained with calcein/cobalt and mitotracker orange .  Calcein labels cytoplasmic as well as mitochondrial regions within cells.  It has been reported that you can specifically identify mitochondria using calcein by quenching the cytoplasmic calcein signal with cobalt (cobalt cannot enter through the mitochondrial membrane).  This technique was reported to specifically localize mitochondria in other cultured cell types.  The cobalt-quenched calcein signal is shown in red.  In green, another mitochondrial specific fluorescent dye, MitoTracker Orange.  We show here that cobalt-quenched calcein only labels a fraction of mitochondria within astrocytes.
